In different healthy and pathological contexts, CD4 and CD8 T cells differentiate into a range of specialized effector and regulatory subsets. In mouse models of aging and autoimmune diseases we study the effector and memory phenotype of CD4 and CD8 T cells. We identified exhausted CD8 T cells with an anti-Ly108 APC antibody in pre-lupus NXBxW F1 mice.

Washed twice in PBS and incubated with L/D fixable aqua dye 20 min at 4°C, then washed twice in PBS.

Surface staining with anti-CD3 BUV805, anti-CD4 BUV495, anti-CD8 FITC, PD-1 PerCP eFluor710, Tim-3 PE Dazzle594, anti-CD62L BV605, anti-Ly108-APC 1/150 etc., in Cell Staining Buffer (Biolegend) incubated 30 min at 4°C.

Different subset of PD-1+Tim-3- exhausted CD8 T cells (Tex), including Ly108+CD62L+ T precursors of exhausted cells (Tpex) could be identified.
