This kit is very helpful to reduce the hands on time when trying to modify a plasmids. It also avoids to re-synthesise/cloning the entire construct. In the other hand, we observe very low efficiency when following the proposed protocol. Longer incubation and plasmid clean-up after PCR amplification partially solve this issue.
