Antibodies are produced by plasma cells (PC) that are terminally differentiated B cells and play crucial roles in host protection against infections but are considered to mediate autoimmunity in patients with rheumatic diseases, such as systemic lupus (SLE) or rheumatoid arthritis (RA). Our research aims at understanding the biology of B cells and antibody-secreting cells (ASC) in health and disease, but in this experiment we are titrating anti-CD2 APC-Vio770.

In this work, we were looking for surface markers that are differentially expressed by B cells (CD19+) and plasma cells (PC). But here we just performed the anti-CD2 titration. CD2 is expressed in CD4 T cells, thus we gated on CD4 to find the best dilution, which is 1/200.
It works and it is affordable.
It is not bright.
The antibody is OK.
