We knocked out an upstream JAK protein (JAK KO) and wanted to examine if there were changes in BIM expression in JAK KO cells. We examined BIM levels in the presence and absence of radiation treatment.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Cal27

Primary Incubation

1:1000 dilution of primary antibody in 1% BSA incubated overnight at 4 degrees celsius on a slow rocker.

Blocking Agent

5% milk

Secondary Incubation

1:1000 goat-anti-rabbit HRP conjugated antibody suspended in 1% milk for one hour at room temperature

Tertiary Incubation

N/A

Detection

Film

Results Summary

We found that there were no significant changes in BIM expression in the JAK KO cells compared to the control cells, regardless of radiation treatment. There are several different isoforms of BIM. The larges isoform is the only one that is clearly visualized by the antibody.

Summary

The Good

Goosd visualisation of the largest isoform of BIM.

The Bad

The smaller isoforms of BIM are barely visible on the blot.

The Bottom Line

This is a great antibody for visualizing the largest isoform of BIM, but a different antibody may be preferred if you are trying to examine the smaller isoforms.
