I seeded the epithelial cells for 24 hours. I starved the cells for 4 hours (0% FBS) and I induced for 24 hours with PDFG and treated them simultaneously with eiF4E siRNA in 2% FBS. I got a clear band. I suggest using low voltage during the western due to the high molecular weight of the protein.
