Mouse spleen, lymph node
Block: 1:50 in 100uL at 37C for 30 min
Fc Block (aCD16/32)
L/D: Zombie NIR 1:4000 in 100 uL at RT for 20 min
Surface stain: 1:200 in 100uL at 4C for 30 min
Spectral flow cytometry (Cytek Aurora)
Very poor staining of NK cells, really only useful as part of a dump gate to partially exclude NK cells from analysis. Difficult to cleanly separate Nkp46+ NK cells from non-NK cells, some NK cells will definitely bleed into the negative gate.
Can be used in a dump gate.
Not very bright, requires UV laser.
Not a very good antibody for flow cytometry.
