CD16 is expressed on granulocytes and on NK cells (lymphocytes). Although the identification of grans is relative easy using a FSC/SSC plot, there are occasions when a good granulocytic marker is needed (e.g. in MDS). Also, NK cell identification is better when both CD56 and CD16 are included. We got this antibody from Beckman to help in both the above tasks.

Results Summary

The results show excellent separation of grans and NK cells from other blood cells. This was achieved even with only 2uL per test! The antibody binds well, and has a bright signal (as shown in the histogram). One needs to be careful though about spillage into neighbouring channels, and tandem breakdown. Keep an eye on the signal in the APC channel.

Summary

The Good

Strong signal. Only 2uL per test is enough.

The Bad

Expensive! Spills into APC.

The Bottom Line

This is a good, reliable antibody. It is ASR, so that adds to the quality and reliability. But it comes at a price. Coulter charge too much for their ASR antibodies.
