I regularly use these tablets for the substrate buffer preparation for the ELISA. The SIGMAFAST OPD (o-phenylenediamine dihydrochloride) tablet set is ready to use tablets to prepare the substrate buffer by dissolving in 20 ml volume of water to detect peroxidase activity in an ELISA assay. Always use freshly prepared substrate buffer using these tablets just before the assay. One set of tablets are sufficient to do four 96 well plates.

Results Summary

A good yellow color gradient was observed within 5 minutes of the substrate buffer addition and the reaction was stopped using 3 M HCl after 15 minutes of incubation.
