NK cells are very important in bridging the innate and adaptive immune systems. They typically either express CD56 dimly with bright CD16, or brightly with dim/negative CD16. Using CD56 antibody to identify them is the best way, and for that we needed a good CD56 antibody that works, and as a bonus can differentiate between 56dim and 56bright cells.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Human peripheral blood

Primary Incubation

10 minutes at RT in the dark

Blocking Agent

1% foetal bovine serum

Secondary Incubation

None

Tertiary Incubation

None

Detection

FL-7 channel on a Coulter Navios cytometer

Results Summary

The results were better than expected. The antibody helped delineate NK cells, and also differentiate between CD56dim NK cells and their less mature counterparts CD56bright cells. The signal was bright enough to show two populations (see figure); the small population at the right end is the CD56bright NK cells, the one in the middle shows the 56dim NK cells, and the large population on the left are the non-NK cells (B, T, other lymphocytes).

Summary

The Good

Easy to use, strong signal

The Bad

Expensive, need relatively large vol/test

The Bottom Line

This is a good antibody from Coulter. It identifies NK cells clearly on a channel that is not always easy to use.
