Be very patient when layering different densities of percoll into your gradient, adding layers down the side of the tube is the best way to avoid mining the layers
When creating a three layer percoll gradient for mTEC isolation from the thymus using this product, we are consistently able to recover 3-4 million epithelial cells per mouse with good clean separation of the thymocytes
