Each tube of compensation beads is stained with one antibody from the staining panel. Create as many tubes of single-stained beads as needed to include all colors that you are staining with

Experimental Design and Results Summary

Application

Flow cytometry

Starting Material

Compensation beads and conjugated antibody

Tips

Bottle must be vortexed well before use each time, and gate out clumps of beads as needed to allow for gating on clean peaks

Results Summary

Compensation beads are stained with one fluorescent antibody in a FACS tube following the same staining procedure as for samples. Create as many tubes as needed to account for all stains in the panel. After removing doublet beads/debris by gating, these beads demonstrate a positive peak for beads that captured antibody and a negative peak which does not bind antibody. You may separately gate on these unstained peaks to use as the negative for each color, or use a tube of unstained beads as a universal negative.

Summary

The Good

Consistent and reliable fluorescence in all channels. This product will bind any mouse, rat, or hamster antibody

The Bad

Aggregates of beads are common but can easily be excluded by gating, leaving only the clean peak of single beads

The Bottom Line

Must-have product for routine flow cytometry users. It is a major advantage not needed to reserve cells for generating single stained compensations
