Cells grown to 80% confluency in 35-mm cell culture dishes are washed with 1×PBS and lysed in ice-cold 100 μl RIPA buffer containing 1 μl protease inhibitor cocktail. Supernatant from cell lysates is used for protein estimation, gel electrophoresis, and western blotting.
