I used this antibody to determine the levels of PTEN in 293T cells post transfection with siRNA against Akt.
Western Blot
Scramble siRNA (first lane from left) and Akt specific siRNA-transfected 293T cells (middle and last lane to the right)
1:1000 dilution, 18 hours at 4 degrees with gentle shaking
5% non-fat dry milk in Tris buffer containing 0.1% Tween-20
HRP-conjugated anti-rabbit antibody 1:2000 dilution, 1 hour at room temperature with gentle shaking

PTEN gets upregulated in response to Akt knock down.
