We bought this neutralization assay with the spike RBD on the plate to test if any of our compounds would block the binding of ACE2 to Spike RBD. The kit was easy to use and experimental results were reproducible.

Tips

Follow the instructions for storage once you have made the detection solutions. They send several detection antibodies that are made into stock solutions and diluted further for use in the ELISA, but these stock solutions have different storage directions than the initial reagent.

Results Summary

We repeated our experiment on different days and were able to reproduce our results. The pop-out well strips make it easy to only use part of the plate each time. The assay does not state the concentration of ACE2 that is added, so you cannot get quantitative results for any inhibition. You can look at the concentration curve of expected binding of ACE2 to the Spike RBD on the plate and based on absorbance have a rough estimate.

Summary

The Good

Everything is included. ACE2 and detection reagents are separated into 2 aliquots to make running multiple experiments easier. Good instructions. Reproducible results.

The Bad

No knowledge of ACE2 stock solution concentration. Some stock solutions are listed to only last 1 day at 4C, so take care to store them properly.

The Bottom Line

Overall, the ELISA neutralization assay kit worked well for our needs and gave reproducible results. We used it within the first week of arrival and stored reagents as suggested, so I cannot comment on the long term stability of the plates/reagents.
