Our lab is interested in looking at the levels of AMPK-a in comparison to the phosphorylation of AMPKa at the Thr172 site. Ny looking at the total amount, we are curious about whether there is a difference in total AMPK-a if we activate T cells in different conditions that changes the downstream mTOR signaling pathway.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Activated murine CD8+ T cells

Primary Incubation

1:5000 in 4 deg overnight

Blocking Agent

5% nonfat dry milk

Secondary Incubation

Anti-rabbit IgG HRP-linked

Tertiary Incubation

N/A

Detection

ECL Pico

Results Summary

The antibody works consistently, showing that the levels remain almost the same during different conditions which is good for getting a ratio of the phospho-ampk and total ampk.

Summary

The Good

Consistency is the key for all experiments therefore this gives very consistent results

The Bad

None

The Bottom Line

Cell Signaling always give promising results and consistency and it carries over throughout their antibodies.
