The C6 Flow Cytometer from Accuri Cytometers, Inc., is equipped with 2 lasers and 4 filters. Together with forward and side scatter, it can perform multiple color analysis in a single cytometry assay, which makes it feasible to perform cell counting and multiplexing, etc. The “analyze and statistic” function within the C-Flow software enables users to interpret data easily. On the downside, however, the C6 is a single channel cytometer without an automated sampler to deliver samples from a microtiter plate. In order to improve throughput, the system can be connected to an automated sampler. The simplicity of the C6 hardware makes this connection easy.

I used the C6 to count approximately 60 samples of high density CHO cell culture. Typically, I stained the cells with 7-ADD. I used the fluorescent detector to detect the dead cells and used forward scattering to detect live cells. The system gave us consistent and accurate results. I periodically compared the results with those using hemocytometer. The results were always within a reasonable range of one another. I also enjoy the fact that the C6 automatically cleans itself when you turn it off, so that I don’t have to decontaminate every day after use.

When I first started using the C-Flow software that interfaces with the C6 Cytometer hardware, I found it difficult to locate the population of interest. This is because the scale of the axes are so large that you must zoom in multiple times in order to see your cells. However, once you get used to it, it is becomes a much easier task.

Overall, the C6 is an affordable flow cytometer that can perform cytometry analysis, including accurate cell counting. It is a low maintenance instrument. Although the software is not user friendly, the user can become proficient with hands-on experience. The problem with throughput can be resolved by integrating the C6 with an autosampler.

The Good

Affordable! This is the least expensive cytometer I have found. It can perform most of the cytometer tasks. It is reliable. Analytical and statistical software are included. No additional software is needed.

The Bad

It is a single channel cytometer, thus throughput is low. Moreover, its software is not easy to use. The scales on the window are too wide to locate the cell population. It takes some time to get use to the C-flow software.

The Bottom Line

An affordable system that can provide what you need from a cytometer.
