We are investigating the used Tobacco mosaic virus (TMV) particles as vaccine candidates. We sought to determine how TMV is taken up in antigen presenting and other immune cells, using fluorescent TMV.

The coupling reaction we used was very similar to those used with labelling antibodies with FITC. Purification was performed via PEG precipitation. It is important to spin the labelled protein for 15000 g for 1-2 mins to pellet any aggregated debris. This gives less background when viewing cells.
TMV particles were brightly labelled and easily visualized upon fluorescence microscopy. The particulate uptake of TMV revealed endosomal compartmentalization.
