My general research aim is to investigate the role of autophagy in neurodegenerative and infectious diseases, using stem cells and stem cell-derived human cellular platforms. For this experiment, we wanted to detect the ratio between ADP and ATP in knockout and control cells. We used this kit as other BioVision kits we have used in the past were useful and sensitive.

Set up the empty plate in the morning to burn residual ATP for a few hours (we left it for 5 hours)

Results Summary

The kit worked beautifully the first time we used it. The results were generated as luminescence values from each well. After measuring ATP, we added the ADP converting enzyme to obtain the ADP values.

Summary

The Good

Very sensitive kit, does not need lots of cells to detect ATP and ADP

The Bad

There is no protein quantification in the process, so you can only obtain % values for the ratio of ADP/ATP.

The Bottom Line

Very good kit although the instructions in the data sheet were not that clear. We didn't grasp that we need to transfer 50 ul of cells from step 5 into the empty plate from step 3. However, the tech support of the company was very responsive and helpful when we wanted to clarify this.
