Our lab is interested in studying the role of inflammatory kinases as a therapeutic target. We perform ELISA and western blot assays to determine the role and pro-inflammatory status of macrophages in response to pharmacological inhibition of these kinases. This kit was good to use and contained easy controls to use. I used this to quantify NO from macrophages in culture and response to LPS.

Protocol Overview

RAW murine macrophages were stimulated and treated with a TAK1 inhibitor at varying concentrations. The cells were then tested for NO production using this kit. The instructions were easy to follow.

Tips

Results Summary

This was an easy way to monitor NO production. The kit worked well. The only down fall is there is not the inclusion of a standard curve to understand the amount of NO being made. Rather you can show RFU values.
