HuR is a stabilizing RNA-binding protein that is encoded by the ELAV-like RNA-binding protein 1. It contains 3 RRMs (RNA recognition motifs) which selectively bind to AU-rich elements (AREs) present in the 3' untranslated region of messenger RNA. It shuttles between the cytoplasm and nucleus, though predominates in the latter. In our lab we utilize it to obtain images of HuR KO and wildtype cells to validate the KO.
Immunohistochemistry
Primary Microglial Cells
Mouse HuR 1:300 in blocking buffer
1% BSA 0.2% milk in PBST
Anti-mouse donkey cy3 1:1000
Hoescht 1:10000
Immunofluoresence Confocal Microscopy
It can be observed that the HuR is localized in the nucleus, due to the overlap with the nuclear stain. In KO there is complete absence of the staining.
N/A
It is highly specific and sensitive
It needs optimization as it can leach into other channels
Optimize and identify the dilution for each different experiment.
