In order to extract out live or dead cells while running flow cytometry, we use this dye to stain for live cells and gate out the cells that may be dead in order to give out a more accurate signal. The dye is very bright and works well.

Stain with surface stain, before fix/perm, in 4C and in the dark.

Tips

We like to aliquot small eppendorf of the dye to make it easier to freeze-thae everytime.

Results Summary

If following the protocol, the stain should be extremely bright and it would be extremely easy to see the two populations. By staining the live cells only, it makes analyzing our data much more accurate.

The Good

Very bright and easy to use in 1:5000 dilution

The Bad

It is light sensitive so always work with foil, cover, or lights dimmed.

The Bottom Line

Definitely a must for flow cytometry if you want to gate out the dead cells in your samples. Due to the nature of the procedure, dead cells are inevitable; therefore, this is useful for staining the live cells only.
