BD Horizon™ BV421 Hamster Anti-Mouse CD120b (TNFR2)

Catalog Number:

CD120b is a receptor for TNF alpha and is also known as TNF receptor 2 (TNFR2). To track expression of TNFR2 on neutrophils, I stained with anti-CD120b-BV421. I titrated the antibody in a surface stain containing markers to stain for neutrophils. Anti-CD120b stained cells well in the range of 1:100–1:200 (0.5–1.0 µL per stain). The antibody works well when used with FMO.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Murine neutrophils (derived from progenitor cells in vitro)

Primary Incubation

Antibodies at indicated amounts (other antibodies for neutrophil surface markers also included to gate neutrophils)

Blocking Agent

FcBlock and BSA were present in staining cocktail. BSA was present in wash buffers.

Secondary Incubation

N/A

Tertiary Incubation

N/A

Detection

Fluorescence by flow cytometry

Results Summary

While it wasn't possible to easily distinguish positive and negative TNFR-expressing populations, a population of neutrophils that stained positive for TNFR2 was easily seen with a fluorescence minus one (FMO) staining control. There wasn't much difference in the staining pattern between 1:200 dilution (0.5 ul/sample) and 1:50 dilution (2 ul/sample).

Summary

The Good

Stains well to see TNFR2 staining

The Bad

FMO absolutely required to distinguish positive and negative populations

The Bottom Line

This reagent works well for application where expression of TNFR2 is tested.
