Our lab is interested in isolating CD8 T cells after harvesting spleens from mice. After isolation, we wanted to look at the CD8 T cell gene expression under varying conditions of glutamine concentration. The isolation was reliable at times but not clean other times.

Protocol Overview

After harvesting the spleens, we grind the spleen against a nylon mesh. Then we follow the manufacturer's protocol to isolate CD8s.

Keep everything on ice

Results Summary

The isolation was reliable at times but also not clean at other times. Our yield was about 90% CD8s. We left the antibodies for longer than the protocol recommended because we think maybe the binding isn't as efficient as before. It worked better after we left it longer.
