We aimed to study the affect of knocking down Setdb1 in mouse prostate cells and change in any methylation pattern. Setdb1 was included as one of the demethylases we aimed to study.

Protocol Overview

The product, siRNA was used with the transfection reagent provided by Dharmacon, according to manufacturer's protocol. Cells were incubated from 72-96 hrs prior to detection of knockdown with western blot.

Tips

Don't overdose the cells. it works wonderfully at concentration mentioned by the manufacturer

Results Summary

About 90% of protein knockdown was observed, however, no changes in methylation were detected.
