We use LUNA-FL for counting and measuring cell viabilities prior submitting the cells to single cell RNA-seq analysis. LUNA-FL was chosen because of it's good capability to count cells and also determine the cell viabilities. Additinally image and report export is easy with LUNA-FL.

Acridine Orange/Propidium Iodide Stain for Cell Count and Viability

Protocol Overview

2 µl of the stain is mixed with 18 µl of the cell suspension. Cells with stain are loaded to PhotonSlide and imaged using LUNA-FL.

Tips

Some cells start to die in the chip, good to do the counting as fast as possible.

Data saving takes a bit long, slides (especially the bright field slides) have some stripes (don't affect the counting)

The LUNA-FL Dual Fluorescence Cell Counter is easy to use and produces fast and reliable results. We have found it to be very helpful in performing fast quality control prior single cell RNA-seq analysis.
