Our goal was to study the stability of H2A in the nucleosomes. For that, we immunoprecipitated Flag-H2A nucleosomes from yeast cell lysates and washed the beads with different NaCl concentration buffers. The IPed nucleosomes were eluted from beads and ran on 15% SDS-PAGE gel and performed western blot with H2A antibody
