Matrix metalloproteinases (MMPs) are important enzymes involved in many physiological processes, including matrix remodeling and cancer invasion. Among MMPs, MMP-9 has been implicated in the induction of tumor angiogenesis and therefore, it is an important mediator of cancer cell dissemination via newly-formed blood vessels. We analyzed the levels of MMP-9 produced by tumor cells and different types of inflammatory leukocytes. To determine the levels of secreted MMP-9, we employed the mouse monoclonal antibody (mAb) 7-11C, available from Santa Cruz (sc-13520). This mAb nicely detects human MMP-9 proenzyme, both as a recombinant protein as well as a zymogen naturally produced by the cells.

In our hands, the mAb 7-11C nicely and reliably detects human MMP-9 proenzyme in western blot assays under non-reducing and reducing conditions.
The mAb 7-11C is also known to prevent the proteolytic processing of MMP-9 zymohen to the activated enzyme. Since this antibody has been raised against MMP-9 proenzyme it might not recognize the activated MMP-9.
The antibody is sensitive and specific as it does not produce any non-specific bands in western blots.
