I have used ThermoFisher's SYBR Safe DNA gel stain to visualize my PCR products used for cloning. PCR products visualized with this stain and gel purified and used for cloning gave improved cloning efficiency with no mutations in the final cloning constructs. SYBR Safe DNA gel stain used was less toxic and safer to use compared to ethidium bromide.

5 microliter of DNA per well in 1 percent agarose gel stained with SYBR Safe DNA gel stain

Protocol Overview

Protocol very simple. Prepare 1% agarose gel in 1X strength TAE buffer. Once agarose gets melted, cool it to 60 degree C. Add SYBR Safe DNA stain to the agarose at 1 in 10,000 dilution. Allow the agarose to set at room temperature (solidification). Once agarose was set, place the agarose in a gel tank and load the wells with DNA marker/ladder and your samples. Perform electrophoresis at required electric volts. Then visualize the gel in BioRad gel imager.

Tips

Protect the dye from exposure to light when it is stored at room temperature.

Results Summary

Very sensitive DNA gel stain. Clear DNA bands were even visualized in Blue Light LED transilluminator.
