Dendritic cells must present intracellular peptides to T cells to induce adaptive immune responses. The cDC1 subset of dendritic cells has been characterized in both mice and humans and specializes in the activation of CD8+ T cells for responses to intracellular pathogens. Key surface markers, including CD24 define cDC1 lineages. We are interested in the development of cDC1s from progenitor cells. This CD24 antibody was selected for use in a multi-color flow cytometry panel for the detection of several DC subsets in different genetic backgrounds.

Results Summary

Dendritic cells were differentiated in vitro from bone marrow progenitors using the cytokine Flt3L. At day 9 of culture when mature DCs have differentiated from multipotent progenitors, cells were harvested and stained for flow cytometry analysis of subsets in the context of different drug treatments. Amongst CD11c+ DCs, a CD24+ population could be readily detected with excellent separation.
