An efficient cDNA conversion is highly required for longer transcripts. Superscript III is highly useful in the conversion of longer transcripts. Superscript III reverse transcriptase is the recombinant MMLV reverse transcriptase with highly reduced RNAse H activity and is a high thermal stability enzyme.

Protocol Overview

5 microgram of the total RNA is used as starting material. The RNA sample is incubated with dNTPS and oligo dt primer at 65 degrees for minutes. Immediately 5X RT buffer and Superscript III reverse transcriptase is added to the RNA sample and incubates at 50 degrees for one hour.

A good quality RNA should be used for good outcome.

Results Summary

Good quality cDNA is obtained which are as long as 12kb in length.
