Pan T cells were isolated from mouse spleen and cultured in vitro for 3 days with IL-2 and DCs. After 3 days, cells were stained with surface markers including CD3, CD4, and CD8. BB700 CD8a was selected because it is significantly brighter as compared to PerCP-Cy5.5. This makes it somewhat easier to compensate against fluorochromes that emit at or near 700nm.
