I use this kit with both injured and uninjured powdered whole lung. I always use the fibrous tissue protocol and generally guesstimate the amount of tissue-typically I get good results eyeballing ~15 ul of powdered tissue and using the largest guideline. From RNA we take it either to RNA seq or just make cDNA and run qPCR. We've gotten excellent results from both.

Tips

After the final 2 minute spin, I always do an additional 2 minutes at top speed with the caps of the filters open to dry everything out; also make sure your sample is fully lysed-sometimes I spin this down twice to be sure it's all clean!

Results Summary

Great ratios for the most part, just make sure the sample is well lysed atthe beginning-I've run into trouble in the past with the lysis step.

Solid kit, but definitely dry the tubes before eluting and make sure your sample is really well lysed-my samples are frozen so this takes some additional vortexing and often an additional spin.
