The Quick Ligation Kit enables ligation of cohesive end or blunt end DNA fragments in 5 minutes at room temperature.
Restriction cloning
Linearized backbone plasmid and insert DNA
Mix 50 ng (0.02 pmol) of backbone and a 37.5 ng (0.06 pmol) of insert with 10 ul of 2x ligation buffer. Add 1 uL of ligase and make up to 20 uL with water. Incubate for 5 minutes at room temperature. Transform into competent cells.
