I’ve tried some other homemade competent cell protocols, but have got higher efficiency cells using the Zymo kit. Additionally, you can skip the heat shock step with these cells.
Cloning
Competent cells & plasmid
Grow up bacteria, do several washes with Zymo wash buffer at 4˚, resuspend in Zymo buffer and freeze at -80˚
Do all steps in cold room at 4˚
Competent cells work great for cloning and transforming plasmids
