Company:

Proteintech Group

Product Name:

phospho(409/410)-TDP43 Antibody

Catalog Number:

22309-1-AP

This antibody works well to detect phospho-TDP43 in samples where there is an abundance of it such as animal or human tissue or lysates. For cell line and primary cell work the signal is dim at best unless you know you have a strong phospho-phenotype. We use this antibody to look at TDP43 phosphorylation status in fractionated tissue lysates and find a ~43kDa band as well as a cleavage band. We have not seen any high molecular weight species.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Tg mouse tissue or primacy cell lysate

Primary Incubation

1:500 in iBind FD Solution

Blocking Agent

iBind FD Buffer for 5minutes

Secondary Incubation

1:10,000 Licor Fluorescent 800 anti-rabbit

Tertiary Incubation

None

Detection

Licor Odyssey Fluorescent imager

Results Summary

Antibody works well as long as you can load enough protein from diseased or stressed tissue/cells to be able to detect significant PTDP signal. We routinely see a full-length and cleavage band using this antibody

Summary

The Good

Works to detect phospho-species via Western Blot and inclusions via staining

The Bad

Need to load a large amount of protein to detect bands on Western Blot

The Bottom Line

Best phospho-antibody we have tried for TDP43 and works in both human and mouse tissues and lysates.
