Our lab examines the interactions of various cell types in the mouse retina. We wanted to be able to separate specific cells from whole tissue samples and then extra RNA from them. These beads allow us to use many different antibodies to capture different cells as needed.

Starting Material

Single-cell suspension from whole mouse retina tissue

Protocol Overview

We digested whole mouse retina tissue and used a single cell preparation protocol to have a solution of all cells from the retina. The beads were pre-incubated with a primary rat anti-mouse antibody (1:50) in DMEM overnight at 4 degrees celcius . After a wash, the beads were added to the sample (50uL per 1.5mL sample) and they were incubated for 1.5 hours at 4 degrees. The tubes were then added to a DynaMag magnet and left to sit for 5 minutes. The supernantent was discarded and the remaining beads+cells were used to extract RNA.

Results Summary

We are able to successfully extract specific F480+ cells from whole cell populations using these beads with low contamination of other negatively selected cell types.
