Ammonium Chloride Potassium lysis buffer is used generally to get rid of the red blood cells from the PBMC populations. Here we have used for performing flowcytometry in PBMC cells.

Protocol Overview

Collect the blood in EDTA coated tube and mix with PBS in the ratio 1:1 and then overlay the blood on to lymphoprep. Centrifuge for 1200rpm and collect the buffy coat and add ACK lysis buffer 10ml and wait for 3-5 minutes and then centifuge again at 300xg for 5 minutes. Discard the supernatant and collect the cells fore futher analysis.

Tips

Slowly discard the supernatant and leave 50ul of the pellet without disturbing the cells.

Results Summary

The cells are free from RBC and it is very good to use for flowcytometry.
