Fisher made a great data sheet for this protocol. It is easy to follow and requires a few simple steps. First you need to know whether you are using a test-tube procedure or a microplate procedure, depending on your method of detection. Additionally, you can perform this assay at three different temperature and times; standard, RT, or enhanced, depending on how much time you have, and if you have access to an incubator or not. They give a simple table for making the standards and working reagent and the settings for proper detection of proteins.
Be aware of the amount of protein you have. To get an accurate concentration reading, youir protein must be in the working range of the assay. Even with cells, we tend to dilute our protein 1:4.
If followed correctly, this procedure is simple and extremely reliable and effective. We usually run our samples in duplicate or triplicate and this assay consistently works well, giving accurate protein concentration readings
