The interaction and communication between the ER and mitochondria has been shown to be involved in development and progression of certain diseases such as obesity and non-alcoholic fatty liver disease. The ability to observe this interaction could aid in determining underlying mechanisms involved in obesity and obesity-related disorders. VDAC is a protein that exists in the outer mitochondrial membrane and has been shown to be involved in interaction with proteins that reside in the ER. The goal of this experiment was to observe VDAC using fluorescent microscopy.

Primary Incubation

1:100 antibody in 1% BSA in PBST for 1 hr. at room temperature

Blocking Agent

1% BSA, 22.52 mg/mL glycine in PBST (PBS+ 0.1% Tween 20) for 30 min at room temperature

Secondary Incubation

1:200 Alexa Fluor dye 488 in 1% BSA in PBST for 1 hr. at room temperature in the dark

Tertiary Incubation

Olympus BX63 microscope with FITC filter

Results Summary

VDAC clearly shows up in green and outside of the nucleus.
