Our laboratory often needs to purify specific cell types in order to infect with HIV, or to engineer immune effector cells such as NK cells and CD8 T cells for enhanced anti-HIV activity. We use the bead-based magnetic separation and the autoMACS machine for this purpose.
Purification of cell types
Mixed population of cells, such as human PBMC or mouse splenocytes
We use Miltenyi beads to capture the cell surface markers of interest. Cells are incubated at the specified dilution in the bead kit for 15 min at 4 degrees Celcius, then washed with running buffer, spun down at 1200 RPM for 10 min, and resuspended in 2 ml of running buffer in 15 ml conical tubes. Tubes are then loaded on the rack and the separation program is run.
Good maintenance of the machine is required; otherwise parts start breaking down or becoming problematic
The machine is very useful and it gives a good separation of the desired cell populations. It is completely automated, and therefore it becomes very handy when separating from a large starting number of cells. However, the recommended maintenance work are a must. We neglected some of those maintenance visits and we had problems with parts such as the injection needle and one of the valves.
Machine works well, I recommend its use as long as maintenance is not neglected
