We tested this kit to isolated CD8 T cells from human PBMCs. We decided to test this product because it is significantly cheaper than similar kits from different companies.

Protocol Overview

Resuspend PBMCs at 1*10^8/ml. Add 10uL of antibody coctail per 1^10^7 cells. Incubate on ice for 15min. Without washing add 10uL of streptavidin microbeads. Mix and incubate on ice for 15min. Resuspend in 3ml of buffer and place the tube in the magnet for 5min. Wash 3x. Collect unlabeled cells.

Results Summary

We get a reasonable purity of CD8+ T cells (89%). But the yield was very low with a large CD8 positive population (20%) left in the negative cell fraction.

Summary

The Good

High purity of the CD8 positive cell fraction, cells of interest are unlabeled

The Bad

Low yield, cheaper price than similar kits, can not use the negative fraction for further separations.

The Bottom Line

Good kit if you only want to separate the CD8 positive cells that are unlabeled. Not good if you want to take the CD8 negative cells to purify other cell populations. The yield is not great so if you need a high cell number will need to start with more PBMCs and use more reagents.
