The Working Antibody for Versican

Our aim is to find if the TGF-beta superfamily member, Nodal, induces the expression of versican components. Since versican has several isoforms, the current antibodies targeting versican on the market is very tricky. Most of them didn't give us a reasonable result. Some of them only detect a band at 70 KD, which is unlikely a potent component of versican protein. Bosterbio claims this antibody dectects a band over 250 KD. Since versican is a very big protein. It sounds this is a real antibody. After testing, we found we can see a clear detected band around 245 KD. We realized that this might be the true result. The reason we got the lower band is perhaps due to the endogenous processing of the protein, and some part of the protein is cut off. Furthermore, the siRNA of versican treatment resulted in the downregulation of the same band, which confirmed our hypothesis. Generally, this should be an antibody for real detection of the versican. However, the good antibodies for different isoform part of the versican is still expected.
