qPCR and high throughput sequencing

Results Summary

For qPCR, intact mRNA was used for m6A Ab IP. Enriched mRNA was tested by qPCR using primers for m6A-modified genes. The enrichment fold is around 20, not ideal but reasonable. For high-throughput sequencing, mRNA was fragmented before subjected to m6A Ab IP. Enriched mRNA fragments containing m6A is used to construct mRNA-seq libraries for high-throughput sequencing. The detection sensitivity toward m6A-modified gene transcripts is excellent. Metagene shows good transcriptome-wide enrichment. Ideal for m6A-seq in various organisms.
