We are assessing consequences of Aryl hydrocarbon Receptor (AhR) activation in dendritic cells. Specifically for these experiments we were assessing Aryl hydrocarbon Receptor activation in bone marrow derived dendritic cells. Precursor cells are isolated with Lympholyte M from whole bone marrow suspensions and grown in the presence of GM-CSF for 5-7 days. We then use flow cytometry and RT-PCR to assess changes in dendritic cells after AhR activation.

Cells were stained with fluorochrome conjugated antibodies and collected with the FACSAriaII Flow cytometer.
Filter cell suspension immediately prior to collection
The FACSAria II Flow Cytometer is pretty bulky and analysis is relatively slow. The results are clean and consistent and the software is fairly navigable.

Clogs are a common issue with the smaller nozzles.
This machine is great for cell sorting, but slow if flow cytometry is its main use.
