The purpose of this project was to separate soluble proteins from insoluble aggregates in rat liver cells. Before sending samples to proteomics to identify proteins in each fraction, we wanted to be able to visualize each on a polyacrylamide gel to crudely quantify differences in protein concentration in each sample.

Protocol Overview

After electrophoresis has been performed, the gel is rinsed 3 times for 5 minutes. Pour enough simplyblue over the gel to cover it. Shake at room temperature for 1 hour to stain the gel. Wash the gel with water for an hour to overnight for best results.

Tips

Bands are visible without destaining, but bands are much more crisp when destained and look better when imaging.

Results Summary

SimplyBlue successfully stains protein bands bright blue and destaining prevents any bleeding.
