LIVE/DEAD® Fixable Blue Dead Cell Stain Kit, for UV excitation

This dye was used to discriminate between live and dead cells in experiments that needed fixation and permeabilization for intracellular antibody staining. This dye also worked well in experiments that didn't fix and perm but we wanted a viability dye that is not in the PI or 7AAD channel.

Results Summary

The fixable blue dye is excited by the 355nm UV laser on the LSR II and has an emission around 450nm. To minimize background, it is important to use PBS only (no FBS or BSA) during staining. Although the fixable dyes are generally used when the cells are fixed and permeabilized, if you are looking for a viability dye other than PI or 7AAD, you can still use this dye and just not follow through with the fix/perm step. I like Fixable Blue because as of right now, there is no good fluorochrome out in the market for the UV laser (yet to try the BUV395 dye released by BD Biosciences).
