I have used the Merck Millipore columns to concentrate Total RNA. I wanted to maximise RNA recovery by using a number of sample aliquots for the RNA isolatio, but I required high concentration RNA. Therefore, I eluted into multiple eluates, combined them and then concentrated the RNA into a smaller volume using the Merck columns.

Protocol Overview

I loaded each column with 400ul of RNA. I then centrifuged the columns for 88 minutes, inverted the columns in a new tube, and then centrifuged them again for a further 2 minutes to elute the RNA.

Tips

The centrifugation time requires some optimisation

Results Summary

The Merck columns effectively concentrated my RNA into 1/20th of the original volume.
