I work on trying to determine the source of ROS production in eosinophils following the engagement of a eosinophil specific surface receptor. I used this kit to understand the kinetics of ROS following receptor engagements. When looking at potential kits, I decided on this one because it had all the reagents necessary and the protocol was simple.

Protocol Overview

I followed the exact protocol provided by the kit. Cell stimulus was added in normal media, which I washed off with buffer provided by the kit.

Tips

If you are using a cell type that can easily degranulate/activate in tissue culture coated plates, please coat your plates with something like fibronectin. This will save you a lot of time

Results Summary

I used flow cytometry and analyzed the data using geometric MFI. Data can also be analyzed using % ROS production, which can be tricky because the gating can be difficult.
