I was using the secondary antibody to detect a primary HA antibody that was derived from a mouse. The HA should have been nuclear, which is exactly where I found secondary antibody fluorescence. This is a good secondary antibody that detects primary mouse antibodies and gives strong signal for fluorescent microscopy.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Primary Human Epithelial Cells

Primary Incubation

Primary anti-mouse antibody, overnight, 4 degrees

Blocking Agent

3% BSA

Secondary Incubation

Donkey anti-Mouse AlexaFluor 555, 1:500, 1 hour at room temperature

Tertiary Incubation

NA

Detection

Confocal Fluorescent Microscopy

Results Summary

Detected strong 555 signal in primary human epithelial cells stained with a primary mouse antibody against a nuclear localized protein. 555 signal co-localized with nuclear staining indicating it had bound to the primary antibody. There was little non-nuclear signal indicating good specificity.
