This antibody is used to evaluate the number of GAD67+ neurons in various brain regions of mice that have been intoxicated with MPTP or a variety of drugs. Using standard stereological methods it was easy to quantify the neurons using a 40x objective.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Fixed brain tissue

Primary Incubation

1:500 in 2% normal donkey serum/TBS

Blocking Agent

5% NDS/TBS

Secondary Incubation

1:2000 in 2% NDS/TBS, 1 hour, room temperature

Tertiary Incubation

NA

Detection

Texas Red fluroescence

Results Summary

At 40x, it was easy to identify GAD67+ neurons with little background fluorescence or non-specific staining. At lower magnifications (4x) it was difficult to identify the regions where GAD67+ neurons were located.
